Unfortunately, you can't highlight text in Publisher, but you can emphasize text by changing the font color or style.

Select the text that you want to emphasize.
Select the options that work best for you from the Text Box Tools Format tab. Some possible options for highlighting the text are the Font Color, Bold or Italics, and WordArt.
